BioLineRx, Hemispherian to present GLIX1 ovarian cancer data at ESMO

The preclinical data show synergy between GLIX1 and PARP inhibitors in HR-proficient ovarian cancer cell lines.

BioLineRx and Hemispherian will present preclinical data on GLIX1 at the 2026 European Society for Medical Oncology Annual Congress in Madrid.

The accepted e-poster will cover data showing strong synergy between GLIX1 and various PARP inhibitors in homologous recombination-proficient ovarian cancer cell lines.

This is relevant because PARP inhibitors have become an important maintenance treatment option in selected ovarian cancer patients, particularly those with BRCA-mutated or homologous recombination-deficient disease. Their benefit is more limited in HR-proficient tumours, which account for around 50 per cent of high-grade serous ovarian cancers.

GLIX1 is a first-in-class, orally administered, brain-penetrating small molecule activator of the TET2 pathway. BioLineRx and Hemispherian describe the pathway as commonly inhibited in cancer. By activating TET2, GLIX1 is intended to overwhelm cancer-cell DNA repair capacity and trigger apoptotic cell death.

The ESMO 2026 presentation is titled “GLIX1, a TET2 Activator Targeting the DNA Damage Response, Synergizes with PARP Inhibitors in Ovarian Cancer Cell Lines.” It will be presented as e-poster 1435eP.

Hemispherian’s lead programme, GLIX1, is being advanced in partnership with BioLineRx for glioblastoma and a broad range of solid tumours. BioLineRx initiated a Phase 1/2a clinical trial of GLIX1 in the first quarter of 2026.

The ovarian cancer data support a potential route for extending PARP inhibitor utility into patient populations where current benefit is limited. Further clinical work would be needed to determine whether the preclinical synergy can translate into treatment benefit in patients.
